Bioconversion of Corncob to Succinic Acid: Bioprospecting for Succinic Acid Producing Bacteria Within Cow Dung
Short Introductive summary
Therefore, the purpose of this study was to bio-prospect presence of succinic acid (SA) producing microorganisms within cow dung using next generation sequencing (NGS), the potential presence of which will lead to their isolation for further studies. Materials used for this study are one sample of cow dung collected and divided into two sterile plastic bags from a field in Kagiso, a township in Krugersdorp, South Africa. The dung sample was thoroughly mixed in a sterile bag before the division. Qiagen DNeasy PowerSoil kit (BioLabs Inc., New England), 341F and 805R primers (Inqaba Biotec, South Africa), OneTaq 2X Master Mix with Standard Buffer (Inqaba Biotec, South Africa), T100 thermocycler (Biorad, South Africa), Gel Doc EZ imager (BioRad, South Africa), Qubit 2.0 fluorometer (Invitrogen, California, USA), and gel casting plates. A cow dung sample designated for sequencing was stored in a freezer at -4°C to avoid the denaturation of the microbial nucleic acid. The samples were thawed at room temperature before the experiments.
